I think that is not possible.

Hammer Bros. have a start node, but move only with the rules that they do not go to Start Nodes, Towers, Castles, Forks, Mushroom Houses, probably more.

You could still do something simmilar by adding an animated enemy to the tree model of a map

Edit: Also removing star coins from levels is possible.

It's an 3d editor also used alot for animating stuff.

It is paid though and you need a version between 7.0 and 2011 because only these allow installing the SDK plugins. Remember that obtaining these isn't legal and therefore we cannot help you finding these (publically ).

somehow making it where you could leave the level at anytime, similar to the Toad Houses, unless leaving the level at any time is exclusive to Toad Houses.

You can make any entrance go to the current world map node (or a specified world map node) with the entrance setting that is used in toad houses. It doesn't have a box in NSMBe, but you can find use it by hex editing the level.

You said changing star coin values on signs is possible- is moving signs around possible too? Say, in front of a tower?

yes but putting a starcoin sign in front of a tower won't make the level unaccessible if you don't pay, for that you need to make a path to the tower level with the starcoin sign behavior and put the sign on the path

No. The Y axis is the one that goes upwards. Remember that we are dealing with 3d space.

And yes, it is possible. Just do any animations you want in Maya. Support will also be added in Goombatlas if I finally get the animation creator done.
