In case if i every make NSMB Adventure Wii, do xdelta patches work on ISOs and such?

I believe xdelta patches work on anything. However, not sure if it's the best option for something like nsmbwii hacking as I haven't really seen it been used for that.

No, you'd be overwriting the first patch you applied.

Same idea. When you patch using XDelta you are modifying the ROM data, so you'd more than likely just corrupt the ROM.
